Patient snapshot Fictional demo

58-year-old, stage IV colorectal adenocarcinoma (sigmoid primary, resected). Liver and lung metastases. MSS (pMMR), KRAS G12D, BRAF wild-type, HER2 negative. Two prior lines: FOLFOX plus bevacizumab, then FOLFIRI plus bevacizumab, progressed on both. No anti-EGFR therapy (not indicated with a KRAS mutation). ECOG 1, no brain metastases, adequate organ function. Located north of Atlanta; willing to travel 250 miles.

Scope: 123 recruiting interventional colorectal trials within 250 miles, pulled from ClinicalTrials.gov. Top 15 by local site presence analyzed below.

Strong candidate: 1

Pidnarulex + Cemiplimab for refractory MSS colorectal cancer (Phase 1/2) · NCT07147231

Why it matches: built for exactly this situation: MSS colorectal cancer progressed through standard chemotherapy. pMMR/MSS confirmed, metastatic and refractory, measurable disease, and the protocol's required prior progression (fluoropyrimidine, oxaliplatin, irinotecan, bevacizumab) is satisfied, with anti-EGFR waived when contraindicated by a KRAS mutation.

Check before calling, from the NGS report: the Phase 2 cohort requires MYC amplification or an FBXW7 mutation plus a liver lesion. Liver lesion: yes. MYC/FBXW7: check the report; if absent, ask about Phase 1 dose-escalation slots, which do not require the marker.

Drafted coordinator email: the workbench pre-drafts the outreach with a one-paragraph clinical summary and the five screening questions worth asking.

Conditional: 1

Repotrectinib (Phase 1/2) · NCT03093116: relevant only if the NGS report shows an ALK, ROS1, or NTRK1-3 fusion. Rare in colorectal cancer and usually absent with a KRAS mutation, so a long shot, but the check costs one look at a report the patient already has. Nearest site: Athens, GA.

Why phone verification is part of every report

One trial in this very dataset shows overall status RECRUITING while all eight of its southeastern sites are individually SUSPENDED. Another lists a site as COMPLETED under a recruiting banner; a third's only Georgia site is NOT YET RECRUITING. Registry status is a starting point, not the truth. Every strong-candidate site gets a verification call before the patient's oncologist is asked to spend time on it.

Method and scope

Source: ClinicalTrials.gov API, recruiting interventional colorectal trials within 250 miles of Atlanta (123 total). This demo hand-analyzed the top 15 by local site presence; the production workbench evaluates the full set and ranks by fit. Eligibility summaries are plain-English readings of registry criteria; the protocol at the site always governs. This report is informational and is never medical advice.
